Efecto de la salinidad sobre la germinación de cultivares de granos en condiciones in vitro

Resumen

La salinización de los suelos en la actualidad es uno de los problemas más graves que enfrenta la agricultura, ya que afecta significativamente la producción de alimentos; por esta razón se hace necesario el estudio de cultivares tolerantes a estas condiciones para su inclusión a los esquemas productivos del país. El estudio tuvo como objetivo seleccionar por su tolerancia a la salinidad, diferentes cultivares de granos: garbanzo (Cicer arietinum L.); frijol común (Phaseolus vulgaris L.); frijol caupí (Vigna unguiculata (L.) Walp); trigo (Triticum aestivum L.) y guisante (Pisum sativum L.). Se estudió en condiciones in vitro la tolerancia de 15 cultivares de granos frente a diferentes concentraciones de cloruro de sodio (NaCl) (50, 150 y 200 mM), para este fin se determinó: el porcentaje de germinación (PG) y la velocidad de germinación (VG) de las semillas según describe Maguire. De los cultivos estudiados se seleccionaron por su tolerancia a 200 mM de NaCl, los cultivares de frijol PRL-8, Caujerí-2170 y San Francisco-219, de caupí INIFAT-93, INIFAT-94 e INIFAT-202 y de trigo I-399, al mostrar los mejores resultados en cuanto a porcentaje de germinación e índice de velocidad en la germinación, por lo que constituyen materiales promisorios para ser utilizados en agroecosistemas afectados por esta condición. Todas ellos mostraron diferencias en el índice de velocidad de germinación de semillas.

En las últimas décadas, la crisis climática y alimentaria nos ha llevado a replantear nuestra alimentación y sus efectos en el ambiente, ya que el cambio climático provoca una serie de fenómenos desfavorables para el adecuado desarrollo de la agricultura (11. Por otro lado, la escasez de nutrientes fundamentales como nitrógeno (N), fósforo (P) y potasio (K) en los suelos con estas características limita aún más su fertilidad (22. Dentro de ellos la salinización de los suelos constituye uno de los problemas más graves que enfrenta la agricultura, ya que afecta significativamente su productividad, lo que hace difícil la supervivencia de las plantas y otras formas de vida por los altos niveles de conductividad eléctrica, exceso de sales iónicas y bajo potencial hídrico (33. Estos efectos provocan también daños irreparables, que limitan de manera total o parcial el adecuado crecimiento de los cultivos (44. Se estiman que existen aproximadamente 830 millones de hectáreas (ha) con problemas de salinización, lo que es equivalente a más del 6% del área total mundial y aproximadamente el 20% del área cultivable total (55. En Cuba, el 14,9 % de la superficie agrícola está afectada por la salinidad y sodicidad, de ellas el 15 % o más del área bajo riego corre el peligro de salinizarse (77. La Habana: ONEI; 2021.), por lo que el estrés salino se considera una amenaza creciente para el desarrollo agrícola del país.

Una solución parcial a los problemas de salinización de los suelos, es el empleo de cultivos y variedades más tolerantes a la salinidad, capaces de activar mecanismos de defensa que les permite mantener su funcionamiento y adaptarse mejor a condiciones adversas (88. Así mismo tiempo, aislar genes responsables de la tolerancia a este estrés y modificar los genomas de las plantas para una mayor productividad en zonas salinas (99. Por esta razón, se emplean criterios de selección de genotipos tolerantes como indicador para su establecimiento en estas condiciones (99. No obstante, el trabajo con cultivares con estas características en condiciones de campo es una tarea compleja y costosa, por los niveles de semillas a emplear para este fin. Sin embargo, evaluar la respuesta de cultivares desde los primeros estadios de desarrollo, permite mayor efectividad para realizar una selección de los mismos y sus índices de tolerancia al estrés salino (1212. En concordancia con todos estos fenómenos, está la necesidad de transitar hacia dietas más saludables, diversificadas y sostenibles relacionadas con los sistemas actuales de producción y consumo de alimentos, cada vez más creciente. En este empeño los granos juegan un papel clave por su aporte de energía y nutrientes a nivel global y los beneficios para la salud (1313. Hervert HD. Por todos estos aspectos el presente estudio tiene como objetivo seleccionar por su tolerancia a la salinidad diferentes cultivares de granos: garbanzo (Cicer arietinum L.); frijol común (Phaseolus vulgaris L.); frijol caupi (Vigna unguiculata L); trigo (Triticum aestivum L.) y guisante (Pisum sativum L.), para su inclusión en programas de producción de alimentos en el país en áreas afectadas por este tipo de estrés.

Materiales y métodos

 

Material biológico

 

En el estudio se utilizaron 15 cultivares: Garbanzo (Cicer arietinum L.): Nacional-29 (N-29), Nacional 5-HA (N5-HA), Nacional 4x2 (4x2); Frijol común (Phaseolus vulgaris L.): PRL-8, Caujeri 2170, San Francisco-219; Caupi (Vigna unguiculata (L.) Walp: INIFAT-93, INIFAT-94, INIFAT-202; Trigo (Triticum aestivum L.): Cuba C 204, I 399, RM 04, RM 27 y Guisante (Pisum sativum L): Agro C-38, Afila 44 y Afila 64. Procedentes del Banco Central de Germoplasma del Instituto de Investigaciones Fundamentales en Agricultura Tropical “Alejandro de Humboldt”, INIFAT.

Ensayo para evaluar tolerancia a diferentes valores de cloruro de sodio (NaCl) bajo condiciones controladas

 

Las semillas de cada uno de los granos utilizados en el experimento se desinfectaron con hipoclorito de sodio al 4 % durante 15 min y se lavaron tres veces con agua destilada estéril. La unidad experimental correspondió a las placas de Petri de vidrio de 140 cm de diámetro y 20 cm de alto, con papel de filtro humedecido con agua destilada sobre el fondo de las placas. Se emplearon 25 semillas (unidad experimental) de cada cultivar en el interior de las placas. Se aplicaron a cada placa 25 mL de distintas soluciones de cloruro de sodio (NaCl) (50, 150 y 200 mM) (milimoles), y un tratamiento Testigo con 0 mM donde solamente se adicionó agua destilada estéril. Las concentraciones de NaCl utilizadas en el estudio (50, 150 y 200 mM) se seleccionaron con base a trabajos previos que describen la respuesta de la germinación a un gradiente de salinidad. Niveles cercanos a 50 mM se asocian con un estrés leve, en el que la germinación de la mayoría de las especies se mantiene por encima del 70 %, permitiendo detectar diferencias sutiles entre genotipos. En cambio, concentraciones de 150 mM se consideran niveles de estrés moderado-alto, donde se registran reducciones marcadas en el porcentaje e índice de velocidad de germinación, pero con datos suficientes para el análisis estadístico de la tolerancia. Valores de 200 mM de NaCl se describen como salinidad severa, en los cuales se observan disminuciones superiores al 80-90 % en la germinación y en el crecimiento inicial de raíz e hipocótilo, así como importantes alteraciones en el balance iónico (incremento de Na⁺ y disminución de K⁺, Ca²⁺ y Mg²⁺), lo que permite evaluar la respuesta bajo condiciones extremas de estrés (1414. Las placas fueron dispuestas siguiendo un arreglo experimental completamente aleatorizado, a una temperatura de 25 °C y una humedad del 80%. Por cada variante se realizaron tres repeticiones.

Se contaron las semillas germinadas diariamente desde su establecimiento hasta la estabilización, con los datos obtenidos se determinó:

Porcentaje de germinación (PG): El experimento se controló durante 15 días y se consideró como semillas germinadas la aparición de la radícula mayor o igual a 2 mm. Diseño experimental y análisis estadístico

 

Los resultados (PG, IVG) fueron sometidos a un análisis de varianza (ANOVA) con pruebas de Rangos Múltiples de Duncan (5 % de probabilidad de error) para detectar las diferencias entre las medias de los tratamientos. Se empleó para ello el Programa STATGRAPHIS Plus versión 5.0.

Resultados y discusión

 

Al analizar los resultados obtenidos, se apreciaron diferencias significativas entre cultivos y especies, con una reducción en los porcentajes de germinación en algunos genotipos a medida que aumentaron los niveles de salinidad. En sentido general en el porcentaje de germinación (PG), en el tratamiento con 0 mM casi todos los cultivares estudiados mostraron un 100 % de germinación, lo que demuestra la calidad de las semillas utilizadas en el ensayo. En el índice de velocidad de germinación (IVG), se destacan los cultivares de frijol: PR-L8 y de trigo: RM-27 con un 90 % en este indicador en el tratamiento con 0 mM.

En la (Figura 1) se muestran los resultados en el cultivo del garbanzo (Cicer arietinum L.), en cuanto a porcentaje de germinación (PG), en el mismo no se aprecian diferencias significativas entre el control y los 50 mM de cloruro de sodio (NaCl), para el cultivar Nacional-29 con respecto al resto, tendencia que se mantiene a las concentraciones de 150-200 mM de NaCl para el mismo. La respuesta Nacional-5HA y 4X2 tuvo un comportamiento similar, aunque con valores más bajos que el Nacional-29 tanto en porcentaje de germinación como en índice de velocidad de germinación para ambos casos.

En el garbanzo se destacó el cultivar Nacional-29 en porcentaje de germinación y el índice de velocidad de germinación (IVG) con respecto al resto, aunque su mejor comportamiento fue a los valores de concentración entre 50-150 mM. Sin embargo, otros autores describen la sensibilidad a la salinidad de este cultivo, lo que afecta negativamente no solo su germinación sino también los procesos reproductivos de este (1717. Se plantea que este estrés disminuye la floración y fructificación, lo que se traduce en afectaciones en el proceso de fertilización del óvulo. No obstante, existen diferencias entre genotipos de garbanzo en la tolerancia a la salinidad (1818. Sobh M, Zargar TB, Basal O, Shehada AOA, Veres S. Available from: https://doi.org/10.15835/nbha53214515. ), aspecto que también se demuestra en los resultados obtenidos en el presente estudio, en las variables evaluadas, lo que deja clara la necesidad de contar con cultivares con estos atributos.

En la (Figura 2) se expresan los resultados alcanzados en el frijol común (Phaseolus vulgaris L.). Al evaluar el porcentaje de germinación (PG), no se aprecian diferencias significativas entre el control y los tres niveles de cloruro de sodio estudiados. No obstante, en el índice de velocidad de germinación (IVG) a 50 mM se destacan los cultivares PRL-8 y Caujeri-2170, aunque, sobresale Caujeri-2170 en los tres niveles de sal estudiados. Al valorar los resultados en frijol se aprecia que los tres cultivares muestran resultados alentadores para continuar estudios.

En este cultivo otros autores encontraron que al emplear sales sódico-alcalinas se limita el crecimiento de la raíz debido a procesos hidrolíticos, donde el Na+ que actúa en el ámbito de meristemos, destruye las células vegetales e inhibe la síntesis de hormonas, lo que afecta el crecimiento de las plantas (1919. Referente a esto, en otros estudios donde se evaluaron concentraciones salinas similares a nuestra investigación en dos cultivares de frijol, se demostró la disminución no solo del porcentaje, sino también de la velocidad de germinación, al incrementar las concentraciones de este elemento (2020. Al evaluar la respuesta a la salinidad inducida con NaCl, sulfato de sodio (Na2SO4) y bicarbonato de sodio (NaHCO3) en frijol en se observó como la biomasa fresca disminuyó con NaHCO3, mientras que el NaCl la incrementó (1919. Estos estudios dan a conocer las disimiles respuestas que se pueden obtener en este cultivo y la importancia de nuevas investigaciones en este campo, lo que se demuestra en las potencialidades de los tres cultivares evaluados y la posible inclusión de estos en suelos muestren afectados por salinidad.

En la Figura 3 en los resultados en el cultivo del caupi, para el porcentaje de germinación (PG), la tendencia fue muy similar a la que se obtuvo en el frijol, ya que no se aprecian diferencias significativas entre los cultivares INIFAT-93, INIFAT-94 e INIFAT-202 lo que demuestra las potencialidades de estos para su estudio frente a diferentes concentraciones salinas. En el índice de velocidad de germinación (IVG) en los niveles de 50-150 mM NaCl se destacan INIFAT-94 e INIFAT-202 sin diferencias estadísticas entre ellos, y consecutivamente INIFAT-93, aunque es digno de resaltar los resultados positivos de los tres cultivares en sentido general. Este tipo de trabajos en condiciones de laboratorio favorece el ahorro de semillas, y permite, además, evaluar diferentes especies en un periodo corto de tiempo como base para su inclusión en sistemas productivos.

Para el caupi, otras investigaciones describen el efecto varietal tanto en el porcentaje de germinación como en el índice de velocidad de germinación o emergencia del cultivo, donde, además, se observan diferencias en el largo de la raíz, aspecto que sugiere la movilización de las sustancias de reservas o traslocación de estas en la etapa de crecimiento del cultivo (2121. En la Figura 4 en los resultados en el cultivo del trigo en el porcentaje de germinación (PG), no se muestran diferencias significativas entre el testigo y 50 mM de NaCl en los cultivares estudiados (Cuba C-204, I-399, RM-04 y RM-27). La misma tendencia se apreció en el nivel 150 y 200 mM con el cultivar I-399, con resultados afines a los encontrados tanto en el testigo como en el nivel 50 mM, lo que demuestra la superioridad de este cultivar con respecto al resto. De igual manera sucede con el índice de velocidad de germinación (IVG) donde se destaca igualmente el mismo cultivar.

Al respecto investigadores evaluaron 400 cultivares de trigo de diferentes especies durante la germinación y estadio de plántulas, en estos estudios hallaron una alta diversidad genética que les permitió seleccionar los que mostraron mayor tolerancia y susceptibilidad a frente a tenores salinos (2222. Así mismo, en otros estudios se sometieron a distintos niveles de salinidad veinte genotipos, lo que permitió comprobar que la exclusión del Na se relaciona con una mejora iónica (relación Na, K y Na/K) así como con los rasgos fisiológicos presentes en diferentes genotipos de trigo bajo estas condiciones de estrés. Algunos de ellos en presencia de altas concentraciones de NaCl manifestaron problemas de crecimiento en la arquitectura de las raíces, los brotes, reducción del área foliar, desarrollo reproductivo y maduración (88. De igual forma, puntualizan como la respuesta a la germinación fue de las variables con mayor grado de diferenciación frente a este tipo de estrés (2323. EL Sabagh A, Islam MS, Skalicky M, Ali Raza M, Singh K, Anwar Hossain M, et al. Estrés salino en trigo (Triticum aestivum L.) en el contexto del cambio climático: estrategias de adaptación y manejo. Igualmente plantean que el estrés salino afecta las relaciones hídricas de las plantas a través del estrés osmótico e hídrico, razón por la cual algunas plantas desarrollan el proceso de ajuste osmótico para conservar la turgencia celular y garantizar el crecimiento, transporte, acumulación y compartimentación de los iones (2121. Available from: http://dx.doi.org/10.22497/arnaldoa.263.26305. ). Todos estos estudios apoyan los resultados obtenidos en la presente investigación y la importancia de realizarlos.

En la Figura 5 en el cultivo del guisante en el porcentaje de germinación (PG), los tres cultivares mostraron excelentes resultados a 50 mM de NaCl, mientras que a 150 mM se destaca el cultivar Afila 44 con más del 70 % y posteriormente Afila 64 con valores más bajos 60 %. En tanto a 200 mM con solamente 40% Afila 44 y Afila 64 muestran la misma tendencia sin diferencias significativas entre ellos, por lo que no se proponen para nuevos estudios.

En los guisantes se describe como al ser expuestos a concentraciones de sal a corto plazo se incrementa el Na⁺ en la savia del xilema en plantas no aclimatadas, pero si están adaptadas reducen este flujo y mantienen mayor K⁺/Na⁺ en xilema, se altera el funcionamiento de PSII y aumenta la generación de ROS en hojas bajo luz, obligando a activar defensas antioxidantes. Estos ajustes coordinados (control de flujo de iones, secuestro de Na⁺, ajuste osmótico y antioxidante) forman parte de los rasgos multifactoriales que determinan la tolerancia o sensibilidad a salinidad en guisantes. En sentido general en granos se plantea que al incrementar los niveles de NaCl se provoca un retardo en la germinación de las semillas, al igual que ocurre en otros indicadores como la longitud de la raíz y la altura de las plantas (44. Refieren igualmente otros estudios como el ión sodio (Na+) es el responsable de la muerte inducida por estrés salino (2525. Available from: https://doi.org/10.32615/ps.2019.169. ) y como las altas concentraciones de Cl- y Na+ en el suelo inhiben el crecimiento de las plantas, ya que afectan la absorción de agua y procesos bioquímicos, como la síntesis de proteínas y la asimilación de dióxido de carbono y nitratos (44. Se explica así mismo, como la alta concentración de estos compuestos en el suelo, el agua y el aire, genera desbalance en los ciclos biogeoquímicos y las cadenas tróficas de las zonas agrícolas, lo que provoca la disminución de la capacidad productiva y la dificultad para controlar plagas, enfermedades y malezas que han generado resistencia (22. Al analizar los resultados obtenidos en sentido general en cuanto a la germinación y el índice de velocidad de germinación de las semillas con los tenores salinos estudiados, se aprecian diferencias entre cultivos y especies, con una reducción en los porcentajes de germinación en algunos genotipos a medida que aumentaron los niveles de salinidad. Lo que nos permitió concluir el efecto negativo que ejercen los altos tenores de sal en el suelo y la importancia de manejar cultivares que logren tolerar estas condiciones para de esta forma continuar estudios y seleccionar los más promisorias para obtener resultados positivos en su inclusión.

Se muestra en la Tabla 1, los cultivares tolerantes frente a los diferentes rangos de salinidad estudiados.

Los resultados obtenidos en la presente investigación constituyen una importante contribución, ya que facilitan la selección de cultivares promisorios para realizar este tipo trabajos en agroecosistemas afectados por salinidad y de esta forma contribuir a incrementar la diversidad de granos para la alimentación y la agricultura en el país.

Conclusiones

 
  1. Existe una alta variabilidad genética entre el germoplasma estudiado en cuanto a su nivel de tolerancia a la salinidad, con diferencias entre el porcentaje de germinación y el índice de velocidad de germinación.

  2. De los 15 cultivares estudiados siete fueron tolerantes a 200 mM la más alta concentración estudiada, seis de ellos a 150 mM y tres a 50 mM de NaCl, lo que denota sus potencialidades para continuar estudios.

Salinity effect on the germination of grain cultivars under in vitro conditions

Abstract

Soil salinization is currently one of the most serious problems facing agriculture, since it significantly affects food production s; for this reason, it is necessary to study cultivars tolerant to these conditions for their inclusion in the country’s production schemes. The study aimed to select different grain cultivars for their tolerance to salinity: chickpea (Cicer arietinum L.); common bean (Phaseolus vulgaris L.); cowpea (Vigna unguiculata (L.)Walp); wheat (Triticum aestivum L.) and pea (Pisum sativum L.). The tolerance of 15 grain cultivars to different concentrations of sodium chloride (NaCl) (50, 150 and 200 mM), was studied under in vitro conditions.For this purpose, the following were determined: the germination percentage (PG) and the germination speed index (IVG) of the seeds as described by the Maguire index. Of the crops studied the bean cultivars PRL-8, Caujerí-2170 and San Francisco-219, of cowpea INIFAT-93, INIFAT-94 and INIFAT- 202 and of wheat I-399 were selected by their tolerance to 200 mM of NaCl, by showing the best results in terms of germination percentage and germination speed index, which is why they constitute promising materials to be used in agroecosystems affected by this condition. All of them showed differences in the seed germination speed index.

Furthermore, the lack of essential nutrients such as nitrogen (N), phosphorus (P), and potassium (K) in soils with these characteristics further limits their fertility (22. Among these, soil salinization constitutes one of the most serious problems facing agriculture, as it significantly affects its productivity, making the survival of plants and other life forms difficult due to high levels of electrical conductivity, excess ionic salts, and low water potential (33. It is estimated that there are approximately 830 million hectares (ha) with salinization problems, which is equivalent to more than 6 % of the total world area and approximately 20 % of the total arable area (55. In Cuba, 14.9 % of the agricultural area is affected by salinity and sodicity, of which 15 % or more of the irrigated area is at risk of becoming saline (77. A partial solution to soil salinization problems is the use of crops and varieties more tolerant to salinity, capable of activating defense mechanisms that allow them to maintain their functioning and better adapt to adverse conditions (88. Mir RA, Logeshwari TR, Aryendu, Soma-sundaram R. Elevated osmolytes accumulation helps in combating NaCl stress causing negative impacts on growth and metabolism of Vigna radiata (L.). At the same time, isolating genes responsible for tolerance to this stress and modifying plant genomes for greater productivity in saline areas (99. Hence the need to provide knowledge on the current situation and perspectives of agriculture oriented towards food production with emphasis on the characterization of productive scenarios (1010. For this reason, selection criteria for tolerant genotypes are used as an indicator for their establishment under these conditions (99. İbrahimova U, Kumari P, Yadav S, Rastogi A, Antala M, Suleymanova Z, Zivcak M, Tahjib-ul-arif M, Hussain S, Abdelhamid M, Hajihashemi S, Yang X, Brestic M. However, working with cultivars with these characteristics under field conditions is a complex and expensive task, due to the levels of seeds required for this purpose. Nevertheless, evaluating the response of cultivars from the early stages of development allows for greater effectiveness in their selection and their salt stress tolerance indices (1212. Rosenzweig C, Elliott J, Deryng D, Ruane AC. Müller C, Arneth A, Boote KJ, Folberth C, Glotter M, Pugh TAM, Schmid E, Stehfest E, Yang H, Jones JW. In line with all these phenomena is the need to transition towards healthier, more diversified, and sustainable diets related to increasingly growing current food production and consumption systems. In this effort, grains play a key role due to their contribution of energy and nutrients globally and their health benefits (1313. Hervert HD. For all these aspects, the present study aims to select, based on their salinity tolerance, different grain cultivars: chickpea (Cicer arietinum L.); common bean (Phaseolus vulgaris L.); cowpea (Vigna unguiculata L.); wheat (Triticum aestivum L.); and pea (Pisum sativum L.), for inclusion in food production programs in the country in areas affected by this type of stress.

Materials and Methods

 

Biological material

 

Fifteen cultivars were used in the study: Chickpea (Cicer arietinum L.): Nacional-29 (N-29), Nacional 5-HA (N5-HA), Nacional 4x2 (4x2); Common bean (Phaseolus vulgaris L.): PRL-8, Caujeri 2170, San Francisco-219; Cowpea (Vigna unguiculata (L.) Walp): INIFAT-93, INIFAT-94, INIFAT-202; Wheat (Triticum aestivum L.): Cuba C 204, I 399, RM 04, RM 27; and Pea (Pisum sativum L.): Agro C-38, Afila 44, and Afila 64. These were obtained from the Central Germplasm Bank of the Instituto de Investigaciones Fundamentales en Agricultura Tropical "Alejandro de Humboldt”, (Alejandro de Humboldt" Institute of Fundamental Research in Tropical Agriculture) INIFAT (according its acronyms in Spanish).

Trial to evaluate tolerance to different sodium chloride (NaCl) concentrations under controlled conditions

 

Seeds of each of the grains used in the experiment were disinfected with 4 % sodium hypochlorite for 15 minutes and washed three times with sterile distilled water. The experimental unit consisted of 140 cm diameter by 20 cm high glass Petri dishes, with filter paper moistened with distilled water placed at the bottom of the dishes. Twenty-five seeds (experimental unit) of each cultivar were placed inside the dishes. To each dish, 25 mL of different sodium chloride (NaCl) solutions (50, 150, and 200 mM) were applied, along with a control treatment (0 mM) where only sterile distilled water was added. The NaCl concentrations used in the study (50, 150, and 200 mM) were selected based on previous studies describing the germination response to a salinity gradient. Levels close to 50 mM are associated with mild stress, where the germination of most species remains above 70 %, allowing the detection of subtle differences between genotypes. In contrast, concentrations of 150 mM are considered moderate-to-high stress levels, where marked reductions in germination percentage and germination speed index are recorded, but sufficient data remain for statistical analysis of tolerance. Values of 200 mM NaCl are described as severe salinity, where decreases exceeding 80-90 % in germination and initial root and hypocotyl growth are observed, as well as significant alterations in ionic balance (increase in Na⁺ and decrease in K⁺, Ca²⁺, and Mg²⁺), allowing the evaluation of the response under extreme stress conditions (1414. The dishes were arranged following a completely randomized experimental design, at a temperature of 25 °C and 80 % humidity. Three replicates were performed per variant.

Germinated seeds were counted daily from establishment until stabilization. The following parameters were determined from the data obtained

Germination Percentage (GP): The experiment was monitored for 15 days, and seeds were considered germinated when the radicle emerged to a length equal to or greater than 2 mm. Experimental design and statistical analysis

 

The results (GP, GS) were subjected to analysis of variance (ANOVA) with Duncan's Multiple Range Test (5 % error probability) to detect differences among treatment means. The STATGRAPHICS Plus version 5.0 software was used for this purpose.

Results and Discussion

 

When analyzing the results obtained, significant differences were observed between crops and species, with a reduction in germination percentages in some genotypes as salinity levels increased. In general terms, regarding germination percentage (GP), almost all cultivars studied showed 100 % germination in the 0 mM treatment, demonstrating the quality of the seeds used in the trial. Regarding the germination speed index (GSI), the bean cultivar PR-L8 and the wheat cultivar RM-27 stood out, achieving 90 % in this indicator in the 0 mM treatment.

Figure 1 shows the results for chickpea (Cicer arietinum L.) in terms of germination percentage (GP). No significant differences were observed between the control and the 50 mM sodium chloride (NaCl) treatment for cultivar Nacional-29 with respect to the others, a trend that remained at 150-200 mM NaCl concentrations for the same cultivar. The response of Nacional-5HA and 4X2 was similar, although with lower values than Nacional-29 in both germination percentage and germination speed index.

In chickpea, cultivar Nacional-29 stood out in germination percentage and germination speed index (GSI) compared to the others, although its best performance was at concentration values between 50-150 mM. However, other authors describe the sensitivity of this crop to salinity, which negatively affects not only its germination but also its reproductive processes (1717. Khan HA, Siddique KHM, Colmer TD. Vegetative and reproductive growth of salt-stressed chickpea are carbon-limited: sucrose infusion at the reproductive stage improves salt tolerance. J. Exp. It is proposed that this stress decreases flowering and fruiting, resulting in effects on the ovule fertilization process. Nevertheless, differences exist among chickpea genotypes in salinity tolerance (1818. Sobh M, Zargar TB, Basal O, Shehada AOA, Veres S. Available from: https://doi.org/10.15835/nbha53214515. ), an aspect also demonstrated in the results obtained in the present study for the evaluated variables, thus clearly indicating the need for cultivars with these attributes.

Figure 2 shows the results obtained for common bean (Phaseolus vulgaris L.). When evaluating germination percentage (GP), no significant differences were observed between the control and the three levels of sodium chloride studied. However, regarding germination speed index (GSI) at 50 mM, the cultivars PRL-8 and Caujeri-2170 stood out, although Caujeri-2170 excelled at all three salt levels studied. When assessing the results in common bean, it is evident that the three cultivars show promising results for further studies.

In this crop, other authors found that the use of sodium‑alkaline salts limits root growth due to hydrolytic processes, where Na⁺ acts on the meristematic regions, destroys plant cells, and inhibits hormone synthesis, thereby affecting plant growth (1919. In this regard, other studies evaluating salt concentrations similar to those used in our research on two bean cultivars demonstrated a decrease not only in germination percentage but also in germination speed as the concentrations of this element increased (2020. Paul WS, Afkairin A, Andales AA, Qian Y, Davis JG. Assessing salinity tolerance in pinto bean varieties: implications for sustainable agriculture. Agronomy. 2024;14:1877. When evaluating the response to salinity induced by NaCl, sodium sulfate (Na₂SO₄), and sodium bicarbonate (NaHCO₃) in common bean, it was observed that fresh biomass decreased with NaHCO₃, whereas NaCl increased it (1919. These studies reveal the diverse responses that can be obtained in this crop and highlight the importance of further research in this field, as demonstrated by the potential of the three evaluated cultivars and their possible inclusion in soils affected by salinity.

Figure 3 shows the results for cowpea. Regarding germination percentage (GP), the trend was very similar to that observed in common bean, as no significant differences were found among cultivars INIFAT‑93, INIFAT‑94, and INIFAT‑202, demonstrating their potential for further study under different salt concentrations. Regarding germination speed index (GSI) at 50-150 mM NaCl, INIFAT‑94 and INIFAT‑202 stood out, with no statistical differences between them, followed by INIFAT‑93. However, the positive results of all three cultivars are noteworthy overall. This type of laboratory study promotes seed saving and also allows the evaluation of different species in a short period of time, serving as a basis for their inclusion in productive systems.

For cowpea, other studies describe varietal effects on both germination percentage and germination or emergence speed index, where differences in root length are also observed, an aspect that suggests the mobilization of reserve substances or their translocation during the crop growth stage (2121. Figure 4 shows the results for wheat. Regarding germination percentage (GP), no significant differences were observed between the control and 50 mM NaCl in the cultivars studied (Cuba C-204, I-399, RM-04, and RM-27). The same trend was observed at the 150 and 200 mM levels for cultivar I-399, with results similar to those found in both the control and the 50 mM level, demonstrating the superiority of this cultivar over the others. The same occurs with the germination speed index (GSI), where the same cultivar also stands out.

In this regard, researchers evaluated 400 wheat cultivars from different species during germination and the seedling stage. In these studies, they found a high genetic diversity that allowed them to select those that showed greater tolerance and susceptibility to saline levels (2222. Zhang R, Wang Y, Hussain S, Yang S, Li R, Liu S, Chen Y, Wei H, Dai Q, Hou H. Study on the effect of salt stress on yield and grain quality among different rice varieties. Front. Likewise, in other studies, twenty genotypes were subjected to different salinity levels, which confirmed that Na⁺ exclusion is related to ionic improvement (Na⁺, K⁺, and Na⁺/K⁺ ratio) as well as to the physiological traits present in different wheat genotypes under these stress conditions. Some of them, in the presence of high NaCl concentrations, manifested growth problems in root architecture, shoots, reduced leaf area, reproductive development, and maturation (88. Mir RA, Logeshwari TR, Aryendu, Soma-sundaram R. Elevated osmolytes accumulation helps in combating NaCl stress causing negative impacts on growth and metabolism of Vigna radiata (L.). Similarly, they point out that the germination response was one of the variables with the highest degree of differentiation under this type of stress (2323. EL Sabagh A, Islam MS, Skalicky M, Ali Raza M, Singh K, Anwar Hossain M, et al. Estrés salino en trigo (Triticum aestivum L.) en el contexto del cambio climático: estrategias de adaptación y manejo. They also state that salt stress affects plant water relations through osmotic and water stress, which is why some plants develop the osmotic adjustment process to maintain cell turgor and ensure growth, transport, accumulation, and compartmentalization of ions (2121. Efecto del estrés salino en el crecimiento y contenido relativo del agua en las variedades IR-43 y amazonas de Oryza sativa "arroz" (Poaceae) Arnaldoa. 2019;26 (3). Available from: http://dx.doi.org/10.22497/arnaldoa.263.26305. ). All these studies support the results obtained in the present investigation and the importance of conducting them.

Figure 5 shows the results for pea. Regarding germination percentage (GP), all three cultivars showed excellent results at 50 mM NaCl, while at 150 mM, cultivar Afila 44 stood out with more than 70 %, followed by Afila 64 with lower values (60 %). At 200 mM, with only 40 %, Afila 44 and Afila 64 showed the same trend with no significant differences between them; therefore, they are not proposed for further studies.

In peas, it is described that when exposed to short-term salt concentrations, Na⁺ increases in the xylem sap of non-acclimated plants, but if they are adapted, they reduce this flow and maintain a higher K⁺/Na⁺ ratio in the xylem. The functioning of PSII is altered, and ROS generation increases in leaves under light, forcing the activation of antioxidant defenses. These coordinated adjustments (control of ion flow, Na⁺ sequestration, osmotic and antioxidant adjustment) are part of the multifactorial traits that determine tolerance or sensitivity to salinity in peas (2424. In general terms, regarding grain crops, it is stated that increasing NaCl levels causes a delay in seed germination, as well as in other indicators such as root length and plant height (44. Chourasia K, More S, Kumar A, Kumar D, Singh B, Bhardwaj V, Kumar A, Das S, Singh R, Zinta G, Tiwari R, Lal M. Salinity responses and tolerance mechanisms in underground vegetable crops: an integrative review. Planta, 2022; 255; 68. Available from: https://doi.org/10.1007/s00425-022-03845-y ). Other studies also report that the sodium ion (Na⁺) is responsible for salt-stress-induced death (2525. Rastogi A, Kovar M, He X, Zivcak M, Kataria S, Kalaji HM, Skalicky M, Ibrahimova UF, Hussain S, Mbarki S. JIP-test as a tool to identify salinity tolerance in sweet sorghum genotypes. Photosynthetica. 2020;58(Suppl 1): 518-528. Available from: https://doi.org/10.32615/ps.2019.169. ) and that high concentrations of Cl⁻ and Na⁺ in the soil inhibit plant growth, as they affect water absorption and biochemical processes such as protein synthesis and the assimilation of carbon dioxide and nitrates (44. Chourasia K, More S, Kumar A, Kumar D, Singh B, Bhardwaj V, Kumar A, Das S, Singh R, Zinta G, Tiwari R, Lal M. All these aspects cause substantial losses in plant growth and development (2626. Ashraf M, Munns R. Evolution of approaches to increase the salt tolerance of crops. It is also explained how the high concentration of these compounds in the soil, water, and air generates an imbalance in the biogeochemical cycles and food chains of agricultural areas, leading to a decrease in productive capacity and difficulty in controlling pests, diseases, and weeds that have developed resistance (22. Martínez SRA, Yáñez DMI, Cantú SI, González RH, Marmolejo MJG. Effects of different land-use systems on soil physicochemical properties of achernozem in northeast México. Studies of this type also demonstrate the role of enzymatic and non-enzymatic defense systems in salinity tolerance in cultivars that managed to grow optimally at 100 mM NaCl (88. When analyzing the overall results obtained regarding seed germination and germination speed index under the studied saline levels, differences were observed between crops and species, with a reduction in germination percentages in some genotypes as salinity levels increased. This allowed us to conclude the negative effect exerted by high salt levels in the soil and the importance of managing cultivars capable of tolerating these conditions, in order to continue studies and select the most promising ones to achieve positive results for their inclusion.

The Table 1 shows the tolerant cultivars against the different salinity ranges studied is presented.

The results obtained in this study represent a significant contribution, as they enable the selection of promising cultivars for use in salinity-affected agroecosystems, thus contributing to increasing grain diversity for food and agriculture in the country.

Los resultados obtenidos en la presente investigación constituyen una importante contribución, ya que facilitan la selección de cultivares promisorios para realizar este tipo trabajos en agroecosistemas afectados por salinidad y de esta forma contribuir a incrementar la diversidad de granos para la alimentación y la agricultura en el país.

Conclusions

 
  1. There is high genetic variability among the germplasm studied regarding their level of tolerance to salinity, with differences in germination percentage and germination speed index.

  2. From 15 cultivars studied, seven were tolerant to 200 mM (the highest concentration studied), six to 150 mM, and three to 50 mM NaCl, indicating their potential for further studies.
