Bioprospección de bacilos asociados a cultivares cubanos de trigo para el control de hongos fitopatógenos

Resumen

El trigo es un cultivo de gran importancia para la alimentación a nivel mundial. En Cuba, es una especie relevante por su consumo y las grandes importaciones que se realizan para cubrir las necesidades del país. Dentro de los factores que afectan sus rendimientos se encuentran los hongos fitopatógenos, los cuales sería conveniente manejar con alternativas biológicas. El objetivo del presente estudio fue caracterizar el potencial antagonista de bacilos Gram positivos procedentes de la rizosfera y el interior de plantas de trigo de los cultivares cubanos ˈCuba C-204ˊ, ˈI 399ˊ y ˈM04ˊ. De los 21 microorganismos aislados, el 95 % liberó enzimas líticas (56 % amilasas, 40 % proteasas, 68 % lipasas y 60 % glucanasas); se destacaron siete por sus resultados para tres enzimas y dos por producir todas ellas. Tres bacterias limitaron el crecimiento de Fusarium sp., 13 incidieron sobre Curvularia sp., cuatro afectaron a Curvularia sp., seis a Bipolaris sp., y cuatro a Alternaria sp, con diferencias en el valor del porcentaje de inhibición entre cepas para cada uno de los patógenos. Dentro de los daños ocasionados a los hongos sobresalen la detención de su crecimiento, el enrollamiento del micelio, la producción de abundantes conidios y la lisis celular. La investigación permitió la selección de los aislados MH2, Mtr2.1, MT3, procedentes del cultivar ˈM04ˈ, y el aislado Crt2.1, del cultivar ˈCuba C-204ˈ, los que se pueden considerar promisorios para nuevos estudios encaminados a la obtención de bioproductos para el control biológico del trigo en Cuba.

Para el control de hongos patógenos en la mayoría de los países se utilizan productos químicos, aun cuando se conoce del daño que estos ocasionan al medio ambiente y al hombre, así como sus efectos en la resistencia de las enfermedades y su bioacumulación (55. Es por ello, que cada día se insiste con mayor fuerza en el uso de métodos más sostenibles que también sean efectivos, como por ejemplo el control biológico, dentro del cual se incluye el uso de microorganismos endófitos y rizosféricos, destacándose el papel que pueden desempeñar géneros como Pseudomonas, Trichoderma y Bacillus, entre otros (55. En Cuba, la producción de trigo no satisface la demanda de la población, lo cual hace que se realicen grandes importaciones del grano y subproductos derivados de su procesamiento (77. En el país, este cultivo no está exento al ataque de hongos fitopatógenos, aspecto que incide negativamente en las producciones. Una de las herramientas que se podrían utilizar para su control son las bacterias promotoras del crecimiento vegetal, las que ejercen su efecto contra hongos patógenos mediante la producción de antibióticos, enzimas extracelulares o toxinas, el parasitismo, la competencia por nutrientes o espacio y la inducción de resistencia sistémica, entre otros mecanismos (88. En este sentido, el aislamiento de bacterias asociadas a la especie vegetal y la selección de aquellas que posean características favorables para el control de patógenos resulta importante, con el fin de generar nuevos productos con efecto biocontrol más eficientes y competitivos. Considerando estos elementos el trabajo se propuso como objetivo caracterizar el potencial antagonista de bacilos Gram positivos aislados desde la rizosfera y el interior de tres cultivares cubanos de trigo, para seleccionar bacterias promisorias para el control biológico en la especie vegetal.

Materiales y Métodos

 

Aislamiento de bacilos esporulados asociados a cultivares cubanos de trigo

 

Se seleccionaron al azar plantas de trigo de 30 días de los cultivares cubanos ˈCuba C-204ˈ (variedad comercial), ˈI 399ˈ y ˈM04ˈ, que se encontraban en el Lote 10 de las áreas agrícolas del Instituto de Investigaciones Fundamentales en Agricultura Tropical “Alejandro de Humboldt”, INIFAT. El suelo en el cual se sembró el material vegetal, clasificado como Ferralítico Rojo Lixiviado (1010 Hernández A, Pérez J.M, Bosch D, Castro, N. Clasificación de los suelos de Cuba. Instituto Nacional de Ciencias Agrícolas e Instituto de Suelos. Mayabeque, Cuba: Ediciones INCA: 2015, 91p. Available from: https://ediciones.inca.edu.cu/index.php/ediciones/index ), tenía un pH neutro, buen contenido de materia orgánica y predominio del calcio sobre el magnesio (Tabla 1). Las semillas en todos los casos procedieron del Banco Central de Germoplasma de Cuba, que radica en la propia institución.

Métodos de análisis: pH (potenciometría); materia orgánica (Walkley y Black); P2O5 (Oniani, por extracción con H2SO4); K, Na (fotometría de llama); Ca y Mg (espectrofotometría de absorción atómica) (1111. Las plantas se colocaron en bolsas de plástico y se trasladaron al Laboratorio de Conservación de Bacterias del INIFAT para su procesamiento. Con el suelo asociado a las raíces se conformó una muestra única por cada cultivar, de la cual se tomaron cinco submuestras de 1 g cada una para el aislamiento de los microorganismos. El material vegetal, se lavó y seguidamente, se separaron las raíces, tallos y hojas. Las fracciones de cada órgano se desinfectaron con hipoclorito de sodio al 4 % durante 5 min y se enjuagaron con agua destilada estéril y se maceraron con la ayuda de un mortero estéril.

Las muestras finales se procesaron según el método de diluciones seriadas (1212. Madigan MT, Bender KS, Buckley DH, Sattley WM, Sthal DA. Brock biology of microorganisms. 15th ed. New York. Las diluciones 10-4 y 10-5 se colocaron a 100 °C de temperatura durante 20 min., y posteriormente, se sembraron en el medio de cultivo agar nutriente. Las placas se incubaron durante 24-48 h a 28 °C. Las bacterias diferentes se purificaron mediante siembras por agotamiento y posteriormente, se realizaron tinciones de Gram y se seleccionaron los bacilos esporulados Gram positivos.

Caracterización de la actividad antagonista de los aislados seleccionados

 

Producción de enzimas líticas

 

Para evaluar la producción por parte de las bacterias de enzimas amilasas, proteasas y lipasas se utilizó el medio de cultivo agar nutriente con almidón soluble (1 %), leche descremada (10 %) y Tween 80 (1 %), respectivamente. En todos los casos el período de incubación fue de 72 h y la temperatura de 30 °C. En las placas con almidón se adicionaron 5 mL de la solución de iodo de Gram para la evaluación de las enzimas amilasas y se consideró como una respuesta positiva la aparición de una zona clara alrededor de la colonia bacteriana. Para las enzimas proteolíticas el criterio de presencia de hidrólisis fue una zona clara alrededor de las colonias en el medio con leche, mientras que para las enzimas lipasas se consideró la formación de un halo en el medio con Tween. Para detectar la presencia de enzimas glucanasas se utilizó un medio con 10 g de carboximetilcelulosa (1313. Herrera A. Manual de medios de cultivo. Editorial Científico Técnica;. 1985. 87 p. Available from: https://biblioteca.casadelacultura.gob.ec/bib/57747 ), el que después del tiempo de incubación se cubrió con una solución de rojo congo (0,1 % en agua destilada) durante 30 min y posteriormente, se lavó dos veces por 5 min con NaCl (1M), contemplando como una respuesta positiva para la liberación de la enzima la aparición de un halo amarillo alrededor de la colonia.

Efecto de las bacterias sobre el crecimiento de diferentes hongos

 

Se evaluaron los siguientes hongos fitopatógenos: Fusarium sp. (cepas 2022), Curvularia sp. (cepa 3579), Curvularia sp. (cepa 3388), Bipolaris sp. (cepa 2856) y Alternaria sp. (cepa 3403), todos conservados en la Colección de Cultivos Puros de Hongos del INIFAT (853 de la Federación Mundial de Colecciones de Cultivos).

En un primer ensayo los aislados bacterianos se inocularon, de dos en dos, cada uno a 20 mm del borde de la placa Petri de 90 mm con medio de cultivo agar papa dextrosa (PDA) (BioCen); y en el centro se colocó un disco de 5 mm del patógeno. Se evaluó la acción de la bacteria sobre el crecimiento del hongo mediante una escala cualitativa, donde: (+) indicó que esta limitó el crecimiento del patógeno y (-) que no ejerció ninguna acción sobre este.

En el segundo experimento, los aislados con resultados positivos se inocularon de forma independiente a 20 mm del borde de la placa y en el extremo opuesto se colocó el disco del hongo. Se midió el diámetro del micelio fúngico tanto en las placas donde se inoculó la bacteria como en placas testigos, donde solamente se inoculó el patógeno. Con los datos obtenidos se calculó el porcentaje de inhibición micelial (IM) como:

Además, se describió el daño ocasionado al hongo mediante observaciones que se realizaron en un microscopio óptico AXIOSKOP 40-Zeiss (40 X de aumento). El periodo de incubación en los dos ensayos osciló entre 7 y 10 días, en dependencia del crecimiento del patógeno (incubadora BINDER, modelo B 28).

En todas las pruebas se utilizaron tres réplicas por cada aislado (igual para los testigos). La temperatura de incubación fue de 28 °C. Para medir las colonias, el halo formado alrededor de estas y el diámetro del micelio fúngico se utilizó un pie de rey mecánico (0,05 cm error).

Diseño experimental y análisis estadístico

 

Los ensayos se realizaron con un diseño completamente aleatorizado. Para el procesamiento estadístico se comprobó la normalidad y homogeneidad de las varianzas (pruebas de Cochran C, Hartley y Bartlett), y se realizaron Análisis de Varianza. Las medias se compararon mediante Pruebas de Rangos Múltiples de Duncan (5 % de probabilidad de error). Se utilizó el programa STATGRAPHICS Plus versión 5.0 (1515. STATGRAPHICS Plus version 5.0 [software]. Herndon, VA: Statgraphics Technologies, Inc.; 2000. ).

Resultados y Discusión

 

Aislamiento de bacilos esporulados asociados a cultivares cubanos de trigo

 

De las muestras de suelo rizosférico y del interior de las plantas de los cultivares cubanos de trigo ˈCuba C-204ˈ, ˈI 399ˈy ˈM04ˈ se aislaron 21 bacterias bacilos esporulados Gram positivos. De ellas, seis procedieron de la rizosfera y 15 fueron endófitas y se distribuyeron en siete aislados de cada cultivar, los cuales fueron seleccionados para el desarrollo de la investigación.

Aunque el agar nutriente es un medio de cultivo rico diseñado para el trabajo con bacterias en general, el calentamiento de la muestra limita la presencia de microorganismos que carezcan de estructuras de resistencia como las endosporas y favorece la selección de representantes de la clase Bacilli (1616. Rodríguez-Hernández MG, Gallegos-Robles MÁ, Rodríguez-Sifuentes L, Fortis-Hernández M, Luna-Ortega JG, González-Salas U. En otros estudios realizados en trigo con los cultivares ˈCuba C-204ˈ e ˈI 399ˈ también se aislaron bacilos Gram positivos esporulados clasificados como representantes de la clase Bacilli (1717. Available from: https://doi.org/10.1016/j.biocontrol.2025.105776.), elemento que indica que es posible encontrar este tipo de microorganismos asociados a cultivares cubanos de la especie vegetal.

Caracterización de la actividad antagonista de los aislados seleccionados

 

La mayoría de los bacilos procedentes desde las muestras relacionadas con los cultivares cubanos de trigo liberaron enzimas líticas, aunque se debe destacar la respuesta de los aislados CTr2-1, CTr3.1, T14 provenientes del cultivar ˈCuba C-204ˈ y de los aislados MH1.1, MT2, MT3, MR3.1, MTr2.1, asociados al cultivar ˈM04ˈ, quienes mostraron resultados favorables para tres de las enzimas evaluadas y en particular, de las bacterias codificadas como ITr2 e IH3.1, aisladas desde el cultivar ˈI 399ˈ, que liberaron todas las enzimas (Tabla 2).

La producción de enzimas líticas es un elemento que se refiere en otras investigaciones para bacilos esporulados Gram positivos, definidos como representantes de la clase Bacilli. Por ejemplo, se ha demostrado para cepas de este grupo la capacidad de liberar enzimas glucanasas, proteasas y amilasas (1919. Rojas-Badía MM, Heydrich-Pérez M, Sánchez-Castro D, Tejera-Hernández B, Ríos-Rocafull Y, Lugo-Moya D. Bacterias del género Bacillus con potencialidades para la sostenibilidad agrícola en Cuba. Se debe destacar, que este tipo de compuestos tiene una acción importante sobre las paredes de los hongos, ya que provoca su debilitamiento o lisis, lo que afecta el crecimiento de los patógenos (2020. Es por ello, que se considera como uno de los mecanismos de biocontrol que presentan las bacterias promotoras del crecimiento vegetal. En este sentido, los resultados alcanzados en la investigación brindan elementos de interés para futuros trabajos de obtención de nuevos bioproductos donde se favorezca la presencia de enzimas con el objetivo de garantizar una mayor efectividad en el control del crecimiento de patógenos fúngicos.

Al enfrentar los aislados ante Fusarium sp. Curvularia sp. Bipolaris sp y Alternaria sp., se comprobó que 13 de las 21 bacterias tenían algún efecto sobre el crecimiento de los hongos, aunque solamente dos de ellas alcanzaron resultados positivos sobre tres patógenos (MT3 y MTr2.1), y solo una (MH2) frente a cuatro de estos. Fue relevante la acción de las bacterias en presencia de la cepa 3579 de Curvularia sp., pues 11 aislados influyeron sobre el hongo (Tabla 3).

Aun cuando las bacterias mostraran efecto antagonista, su acción sobre los hongos fitopatógenos presentó diferencias (Tabla 4).

Medias con letras distintas difieren estadísticamente para cada hongo, según ANOVA con Prueba de Rangos Múltiples de Duncan (5 % de probabilidad de error)

Se puede apreciar en la Tabla 4 como el aislado CTr2.1 se destacó frente a Fusarium sp., mientras que los valores del porcentaje de inhibición para el aislado MT3 fueron altamente significativos en comparación con los otros 10 microorganismos que afectaron a Curvularia sp. (cepa 3579). Para la otra cepa de este género utilizada en la investigación (3388), la mayor respuesta de control se obtuvo con los aislados MTr2.1 y MT3. Este último microorganismo además se destacó frente a Bipolaris sp junto con el aislado MH2. Para Alternaria sp., las cuatro bacterias evaluadas presentaron resultados semejantes. Estas diferencias podrían estar asociadas tanto al patógeno como a la bacteria y la especificidad de los metabolitos que produce con respecto a la especie fúngica presente en la interacción, ya que se apreciaron incluso para hongos pertenecientes al mismo género.

Fusarium agrupa diferentes especies que causan daños a distintos cultivos. Este grupo se incluye dentro de los patógenos que afectan al trigo provocando fusariosis de la espiga, una de las enfermedades fúngicas más importantes para los cereales a nivel mundial (2121. Por su parte, el género Curvularia sp., provoca daños en cultivos como el trigo (22. Błaszczyk L, Salamon S, Mikołajczak K. Fungi inhabiting the wheat endosphere pathogens. 2021 Oct;10(10):1288. Available from: https://doi.org/10.3390/pathogens10101288.) y el arroz (Oyza sativa L.), donde produce manchas foliares y pudriciones de la raíz (2222. Bipolaris y Alternaria se incluyen dentro de los patógenos identificados en semillas de trigo, aunque se conoce que pueden afectar los demás órganos de la planta (22. Los géneros Fusarium y Alternaria además producen micotoxinas (11. Deligeorgakis C, Magro C, Skendi A, Gebrehiwot HH, Valdramidis V, Papageorgiou M. Fungal and toxin contaminants in cereal grains and flours: systematic review and meta-Analysis. Foods. 2023 Dec;12(23):4328. Available from: https://doi.org/10.3390/foods12234328.), metabolitos secundarios que pueden causar serias enfermedades a animales y al ser humano (44. Martín I, Gálvez L, Guasch L, Palmero D. Fungal pathogens and seed storage in the dry state. Plants. 2022 Nov;11(22):3167. Available from: https://doi.org/10.3390/plants11223167.). Por tal razón, su control tiene un valor agregado. Por ejemplo, F. chlamydosporum libera moniliformina, una de las toxinas principales descritas para este género fúngico, mientras que F. graminearum y F. culmorum producen zearalenona, considerada un compuesto carcinogénico. Para Alternaria, por su parte, una de las toxinas más destacadas es el alternariol (11. Deligeorgakis C, Magro C, Skendi A, Gebrehiwot HH, Valdramidis V, Papageorgiou M. Estas referencias ilustran la importancia de establecer estrategias de control para estos géneros fúngicos, donde el uso de medios biológicos tiene gran atractivo por su menor residualidad respecto a compuestos de síntesis química (55. Pandit MA, Kumar J, Gulati S, Bhandari N, Mehta P, Katyal R, et al. Major biological control strategies for plant pathogens. Pathogens. 2022 Feb;11(2),273. Se debe destacar que las bacterias que alcanzaron los resultados más relevantes en cuanto a actividad antifúngica no fueron las más destacadas en la producción de enzimas líticas, ni en el halo de hidrólisis ni en la diversidad de compuestos liberados. En este sentido, existen otros mecanismos descritos para géneros de la clase Bacilli que están involucrados en su acción como biocontrol. Dentro de estos se encuentran la liberación de péptidos de síntesis no ribosomal (NRPs) y policétidos (PKs) que tienen acción antimicrobiana, la producción de hormonas, la capacidad de colonización, la formación de biopelículas y la competencia por espacio y nutrientes, la síntesis de acil homoserin lactonasas (AHSL), la producción de compuestos orgánicos volátiles (VOCs) y la inducción de resistencia sistémica (ISR) (2525. Los péptidos de síntesis no ribosomal producidos por especies de Bacillus como las iturinas, fenacinas, entre otros, han mostrado acción antibacteriana y antifúngica, con efectividad frente a Fusarium, Alternaria, Sclerotium, Cercospora y Colletotrichum. Cepas de B. siamensis presentaron actividad frente a F. oxysporum formea speciales (fsp) cubense (FOCTR4) (2626. Kulkova I, Dobrzyński J, Kowalczyk P, Bełźecki G, Kramkowski K. Available from: https://doi.org/10.3390/ijms24119759.), elemento que se asoció con la producción de una gran variedad de sideróforos por parte de la bacteria (2727. Shen N, Li S, Li SY, Zhang H, Jiang M. The siderophore‑producing bacterium, Bacillus siamensis Gxun‑6, has an antifungal activity against Fusarium oxysporum and promotes the growth of banana. gypt J Biol Pest Control. 2022;32(1):34. Por otra parte, se demostró la acción de cepas de Bacillus sobre C. tuberculata, con valores de porcentaje de inhibición micelial cercanos al 30 % (2828. Toledo-Hernández E, Peña-Chora G, Toribio-Jiménez J, Romero-Ramírez Y, Bolaños-Dircio A, Velázquez-del Valle MG, Hernández-Lauzardo AN, León-Rodríguez R, Silvana Vero S. Isolated rhizobacteria of Jatropha curcas L.: antagonistic activity of phytopathogens and plant growth promoter. Rev Chapingo Ser Cienc For Ambient. 2021;27(2):181-98. Available from: https://doi.org/10.5154/r.rchscfa.2020.04.029.), similar a los alcanzados en el presente estudio para los cuatro aislados seleccionados. Estos autores, relacionaron el efecto inhibitorio con la producción de metabolitos volátiles y difusibles por parte de las bacterias.

Otros estudios destacaron el potencial de Bacillus para el control de Bipolaris sorokiniana, con un alto porcentaje de inhibición para el hongo en condiciones in vitro, así como la presencia en esta bacteria de genes asociados a la actividad antagonista relacionados con la producción de antibióticos, sideróforos, entre otras sustancias (2424. Valenzuela-Ruiz V, Parra-Cota FI, Santoyo G, de los Santos-Villalobos S. Potential biocontrol mechanisms of Bacillus sp. TSO2 against Bipolaris sorokiniana, spot blotch in wheat. ev Mex Fitopatol. 2022;40(2):230-9. Available from: https://doi.org/10.18781/R.MEX.FIT.2201-1.). También se refiere la acción de B. subtilis frente a Alternaria, con resultados in vitro superiores a los alcanzados por plaguicidas comerciales (2929. Zárate-Ramos A, Quero-Carrillo AR, Miranda-Jiménez L, Nava-Díaz C, Robles-Yerena L. Fungicides and Bacillus subtilis against fungi isolated from commercial seed of side oats grama (Bouteloua curtipendula). Rev Mex Fitopatol. 2022;40(1):103-15. Además de los hongos incluidos en el estudio, géneros como Cladosporium y Corynespora también han sido afectados por cepas pertenecientes a la clase Bacilli (1919. Rojas-Badía MM, Heydrich-Pérez M, Sánchez-Castro D, Tejera-Hernández B, Ríos-Rocafull Y, Lugo-Moya D. Bacterias del género Bacillus con potencialidades para la sostenibilidad agrícola en Cuba. Anales de la Academia de Ciencias de Cuba. 2022;12(1). Available from: http://www.revistaccuba.cu/index.php/revacc/article/view/1067. ), lo que demuestra su gran potencial para el control de patógenos fúngicos de alta incidencia en diferentes especies vegetales. Algunas de las sustancias anteriormente referidas podrían haber sido liberadas por los aislados seleccionados en la investigación, lo que explicaría su acción superior con respecto a otros que se destacaron en la producción de enzimas líticas. Diferencias en la concentración de estos metabolitos y su diversidad pueden provocar los distintos resultados alcanzados para cada uno de ellos.

La Tabla 5 resume los principales daños que las bacterias ocasionaron a los patógenos. Nótese que en algunos casos la detención del crecimiento del hongo estuvo asociada a elementos como el enrollamiento del micelio y la producción de abundantes conidios, lo cual podría constituir un criterio de selección con el objetivo de potenciar la acción biocontrol de un producto elaborado a partir de estas bacterias.

Otras investigaciones también comentan el daño que causan especies de Bacillus en la germinación de los conidios, la extensión y morfología del tubo germinativo, la membrana plasmática de los conidios y las hifas, lo cual conduce a la muerte de hongos fitopatógenos como Botrytis cinerea (3030. Destacan igualmente, el papel de compuestos liberados por especies de Bacillus, en particular las fengicinas, en la modificación de la permeabilidad de las membranas de patógenos fúngicos donde se incluye la especie F. graminearum (3131. Ali MA, Ahmed T, Ibrahim E, Rizwan M, Chong KP, Yong JWH. En otros estudios géneros incluidos en la clase Bacilli han mostrado tanto mecanismos indirectos como mecanismos directos de estimulación del crecimiento vegetal. Se ha destacado su potencial para fijar nitrógeno y solubilizar fosfato de calcio, hierro y aluminio, con índices de solubilización entre 1,30 y 2,59 (1919. Rojas-Badía MM, Heydrich-Pérez M, Sánchez-Castro D, Tejera-Hernández B, Ríos-Rocafull Y, Lugo-Moya D. Bacterias del género Bacillus con potencialidades para la sostenibilidad agrícola en Cuba. A ello se une la ventaja desde el punto de vista tecnológico que tiene este grupo de bacterias por su versatilidad metabólica, presentar estructuras que le confieren resistencia ante condiciones de estrés, lo que les permitiría una mayor viabilidad en el suelo una vez que sean aplicados (1414. Considerando los elementos anteriormente mencionados, es un resultado promisorio demostrar que los microorganismos en estudio asociados al cultivo del trigo, posean características favorables para el control biológico, ya que estas podrían permitir contrarrestar el ataque de patógenos y estimular el crecimiento de la especie vegetal mediante la aplicación de bioproductos con un efecto integral sobre la especie vegetal (1919. Rojas-Badía MM, Heydrich-Pérez M, Sánchez-Castro D, Tejera-Hernández B, Ríos-Rocafull Y, Lugo-Moya D. Bacterias del género Bacillus con potencialidades para la sostenibilidad agrícola en Cuba. De los aislados bacterianos se destacaron de manera particular el MTr2.1, el MT3 y el CTr2.1 por su acción significativa frente a los patógenos y el MH2, único que mostró actividad frente a cuatro de las cinco especies fúngicas. Estas bacterias son promisorias para futuros trabajos donde se valore su potencial como principios activos de nuevos controles biológicos para el cultivo del trigo en Cuba.

Conclusiones

 

Asociadas a plantas de los cultivares cubanos de trigo ˈCuba C-204ˈ, ˈI 399ˈ y ˈM04ˈ se pueden encontrar bacterias con potencial para el control de hongos fitopatógenos que afectan esta especie vegetal. En especial los aislados MH2, MTr2.1, MT3 y CTR2.1, seleccionados por su efecto antagonista frente a Fusarium sp., Curvularia sp., Bipolaris sp y Alternaria sp., se consideran promisorios para la formulación de bioproductos destinados al manejo de estos patógenos en Cuba.

Abstract

Wheat is a crop of great importance for global food security. In Cuba, it is a relevant species due to its consumption and the significant imports required to meet the country's needs. Among the factors affecting its yields are phytopathogenic fungi, which would be advisable to manage using biological alternatives. The aim of the present study was to characterize the antagonistic potential of Gram-positive bacilli from the rhizosphere and endosphere of wheat plants of the Cuban cultivars 'Cuba C-204', 'I 399', and 'M04'. Out of the 21 microorganisms isolated, 9 5% released lytic enzymes (56 % amylases, 40 % proteases, 68 % lipases, and 60 % glucanases); seven stood out for their results for three enzymes, and two for producing all of them. Three bacteria inhibited the growth of Fusarium sp., 13 affected Curvularia sp., four impacted Curvularia sp., six affected Bipolaris sp., and four affected Alternaria sp., with differences in the percentage of inhibition values among strains for each pathogen. Among the damages caused to the fungi, the most notable were growth arrest, mycelial coiling, abundant conidia production, and cellular lysis. The research allowed for the selection of isolates MH2, Mtr2.1, and MT3, from the cultivar 'M04', and isolate Crt2.1, from the cultivar 'Cuba C-204', which can be considered promising for further studies aimed at developing bioproducts for the biological control of wheat in Cuba.

One of the factors significantly affecting the yield of this plant species is the attack of pathogens, mainly fungal pathogens (11. For the control of fungal pathogens, chemical products are used in most countries, even though the damage they cause to the environment and to human health is well known, as well as their effects on disease resistance and bioaccumulation (55. Therefore, there is increasing insistence on the use of more sustainable yet effective methods, such as biological control, which includes the use of endophytic and rhizospheric microorganisms, highlighting the potential role of genera such as Pseudomonas, Trichoderma, and Bacillus, among others (55. In Cuba, wheat production does not meet the population's demand, leading to large imports of the grain and by-products derived from its processing (77. In the country, this crop is not exempt from attack by phytopathogenic fungi, an aspect that negatively affects production. One of the tools that could be used for its control are plant growth-promoting bacteria, which exert their effect against fungal pathogens through the production of antibiotics, extracellular enzymes or toxins, parasitism, competition for nutrients or space, and induction of systemic resistance, among other mechanisms (88. In this sense, the isolation of bacteria associated with the plant species and the selection of those with favorable characteristics for pathogen control is important, in order to develop new, more efficient and competitive biocontrol products. Considering these elements, the objective of this work was to characterize the antagonistic potential of Gram-positive bacilli isolated from the rhizosphere and interior of three Cuban wheat cultivars, in order to select promising bacteria for biological control in this plant species.

Materials and Methods

 

Isolation of spore-forming bacilli associated with Cuban wheat cultivars

 

Thirty-day-old wheat plants from the Cuban cultivars 'Cuba C-204' (commercial variety), 'I 399', and 'M04' were randomly selected from Lot 10 of the agricultural areas of the Instituto de Investigaciones Fundamentales en Agricultura Tropical "Alejandro de Humboldt", (Alejandro de Humboldt" Institute of Fundamental Research in Tropical Agriculture) INIFAT (according its acronyms in Spanish). The soil in which the plant material was sown, classified as Leached Red Ferrallitic soil (1010 Hernández A, Pérez J.M, Bosch D, Castro, N. Clasificación de los suelos de Cuba. Instituto Nacional de Ciencias Agrícolas e Instituto de Suelos. Mayabeque, Cuba: Ediciones INCA: 2015, 91p. Available from: https://ediciones.inca.edu.cu/index.php/ediciones/index ), had a neutral pH, good organic matter content, and a predominance of calcium over magnesium (Table 1). In all cases, the seeds were obtained from the Central Germplasm Bank of Cuba, which is located within the same institution.

The plants were placed in plastic bags and transferred to the INIFAT Bacterial Conservation Laboratory for processing. A single composite sample was prepared from the soil associated with the roots for each cultivar, from which five 1 g subsamples were taken for microorganism isolation. The plant material was washed, and then roots, stems, and leaves were separated. Each organ fraction was disinfected with 4 % sodium hypochlorite for 5 minutes, rinsed with sterile distilled water, and macerated using a sterile mortar.

The final samples were processed according to the serial dilution method (1212. Madigan MT, Bender KS, Buckley DH, Sattley WM, Sthal DA. Brock biology of microorganisms. 15th ed. New York. The 10⁻⁴ and 10⁻⁵ dilutions were heated at 100 °C for 20 minutes and subsequently plated on nutrient agar culture medium. The plates were incubated for 24-48 h at 28 °C. Different bacteria were purified by streak plating, and subsequently, Gram staining was performed, and Gram-positive spore-forming bacilli were selected.

Characterization of the antagonistic activity of the selected bacterial isolates

 

Production of lytic enzymes

 

To evaluate the production of amylase, protease, and lipase enzymes by the bacteria, nutrient agar culture medium supplemented with soluble starch (1 %), skim milk (10 %), and Tween 80 (1 %), respectively, was used. In all cases, the incubation period was 72 hours at a temperature of 30 °C. For the starch-containing plates, 5 mL of Gram's iodine solution was added to evaluate amylase enzymes, and the appearance of a clear zone around the bacterial colony was considered a positive response. For proteolytic enzymes, the criterion for hydrolysis was a clear zone around the colonies in the milk-containing medium, while for lipase enzymes, the formation of a halo in the Tween-containing medium was considered positive. To detect the presence of glucanase enzymes, a medium containing 10 g of carboxymethylcellulose (1313. Herrera A. Manual de medios de cultivo. Editorial Científico Técnica;. 1985. 87 p. Available from: https://biblioteca.casadelacultura.gob.ec/bib/57747 ) was used; after the incubation period, the plates were covered with a Congo red solution (0.1 % in distilled water) for 30 minutes and then washed twice for 5 minutes with NaCl (1M). A yellow halo appearing around the colony was considered a positive response for enzyme release.

Effect of bacteria on the growth of different fungi

 

The following phytopathogenic fungi were evaluated: Fusarium sp. (strain 2022), Curvularia sp. (strain 3579), Curvularia sp. (strain 3388), Bipolaris sp. (strain 2856), and Alternaria sp. (strain 3403), all preserved in the INIFAT Pure Fungal Culture Collection (853 of the World Federation for Culture Collections).

In a first assay, bacterial isolates were inoculated in pairs, each 20 mm from the edge of a 90 mm Petri dish containing potato dextrose agar (PDA) (BioCen), and a 5 mm disc of the pathogen was placed in the center. The action of the bacteria on fungal growth was evaluated using a qualitative scale, where (+) indicated that bacterial growth limited pathogen growth and (-) indicated no effect on the pathogen.

In the second experiment, isolates with positive results were inoculated independently 20 mm from the edge of the plate, and the fungal disc was placed on the opposite side. The diameter of the fungal mycelium was measured both in plates where the bacterium was inoculated and in control plates, where only the pathogen was inoculated. The mycelial inhibition percentage (MI) was calculated from the obtained data as:

Additionally, the damage caused to the fungus was described through observations made under an AXIOSKOP 40-Zeiss optical microscope (40× magnification). The incubation period in both assays ranged from 7 to 10 days, depending on the growth rate of the pathogen (BINDER incubator, model B 28).

For all tests, three replicates were used per isolate (same for controls). The incubation temperature was 28 °C. A mechanical Vernier caliper (0.05 cm error) was used to measure colonies, the halo formed around them, and the fungal mycelium diameter.

Experimental design and statistical analysis

 

The assays were conducted using a completely randomized design. For statistical processing, normality and homogeneity of variances were verified (Cochran C, Hartley, and Bartlett tests), and analyses of variance (ANOVA) were performed. Means were compared using Duncan's Multiple Range Test (5 % probability of error). The STATGRAPHICS Plus software, version 5.0 (1515. STATGRAPHICS Plus version 5.0 [software]. Results and Discussion

 

Isolation of sporulated bacilli associated with Cuban wheat cultivars

 

From the rhizospheric soil and internal tissues of the Cuban wheat cultivars 'Cuba C-204', 'I 399', and 'M04', 21 Gram-positive spore-forming bacilli were isolated. Of these, six came from the rhizosphere and 15 were endophytic, distributed as seven isolates from each cultivar, which were selected for further research.

Although nutrient agar is a rich culture medium designed for general bacteriological work, heating the sample limits the presence of microorganisms lacking resistant structures such as endospores and favors the selection of representatives of the class Bacilli (1616. Characterization of the antagonistic activity of the selected isolates

 

Most of the bacilli obtained from samples related to the Cuban wheat cultivars released lytic enzymes. Notably, the isolates CTr2-1, CTr3.1, and T14 from the cultivar 'Cuba C-204', as well as isolates MH1.1, MT2, MT3, MR3.1, and MTr2.1 associated with the cultivar 'M04', showed favorable results for three of the evaluated enzymes. Particularly, the bacteria coded as ITr2 and IH3.1, isolated from the cultivar 'I 399', released all the enzymes evaluated (Table 2).

The production of lytic enzymes is a feature reported in other studies for Gram-positive spore-forming bacilli, defined as representatives of the class Bacilli. For example, strains of this group have been shown to have the ability to release glucanase, protease, and amylase enzymes (1919. Rojas-Badía MM, Heydrich-Pérez M, Sánchez-Castro D, Tejera-Hernández B, Ríos-Rocafull Y, Lugo-Moya D. Bacterias del género Bacillus con potencialidades para la sostenibilidad agrícola en Cuba. It should be noted that this type of compound plays an important role in the fungal cell wall, causing weakening or lysis, which affects pathogen growth (2020. Blanco EL, Castro Y. Therefore, this is considered one of the biocontrol mechanisms exhibited by plant growth-promoting bacteria. In this sense, the results obtained in this study provide valuable information for future work aimed at developing new bioproducts that promote the presence of these enzymes to ensure greater effectiveness in controlling fungal pathogen growth.

When the isolates were confronted with Fusarium sp., Curvularia sp., Bipolaris sp., and Alternaria sp., it was found that 13 out of the 21 bacteria had some effect on fungal growth, although only two of them (MT3 and MTr2.1) achieved positive results against three pathogens, and only one (MH2) was effective against four of them. The action of the bacteria against Curvularia sp. strain 3579 was particularly notable, as 11 isolates influenced the growth of this fungus (Table 3).

Although the bacteria exhibited antagonistic activity, their effectiveness against the phytopathogenic fungi varied (Table 4).

Table 4 shows how isolate CTr2.1 stood out against Fusarium sp., while the mycelial inhibition percentage values for isolate MT3 were highly significant compared to the other 10 microorganisms that affected Curvularia sp. (strain 3579). For the other strain of this genus used in the research (3388), the greatest control response was obtained with isolates MTr2.1 and MT3. This latter microorganism also stood out against Bipolaris sp., together with isolate MH2. For Alternaria sp., the four evaluated bacteria presented similar results. These differences could be associated with both the pathogen and the bacterium, as well as the specificity of the metabolites produced with respect to the fungal species present in the interaction, since differences were observed even for fungi belonging to the same genus.

Fusarium includes different species that cause damage to various crops. This group is among the pathogens affecting wheat, causing Fusarium head blight, one of the most important fungal diseases for cereals worldwide (2121. The genera Fusarium and Alternaria also produce mycotoxins (11. Deligeorgakis C, Magro C, Skendi A, Gebrehiwot HH, Valdramidis V, Papageorgiou M. Fungal and toxin contaminants in cereal grains and flours: systematic review and meta-Analysis. Foods. 2023 Dec;12(23):4328. Available from: https://doi.org/10.3390/foods12234328.), secondary metabolites that can cause serious diseases in animals and humans (44. Martín I, Gálvez L, Guasch L, Palmero D. Fungal pathogens and seed storage in the dry state. Plants. 2022 Nov;11(22):3167. Available from: https://doi.org/10.3390/plants11223167.). For this reason, their control has added value. For example, F. chlamydosporum releases moniliformin, one of the main toxins described for this fungal genus, while F. graminearum and F. culmorum produce zearalenone, considered a carcinogenic compound. For Alternaria, one of the most prominent toxins is alternariol (11. Deligeorgakis C, Magro C, Skendi A, Gebrehiwot HH, Valdramidis V, Papageorgiou M. These references illustrate the importance of establishing control strategies for these fungal genera, where the use of biological means is very attractive due to its lower residue compared to chemically synthesized compounds (55. It should be noted that the bacteria that achieved the most relevant results in terms of antifungal activity were not the most outstanding in lytic enzyme production, neither in the hydrolysis halo nor in the diversity of compounds released. In this sense, other mechanisms described for genera of the class Bacilli are involved in their biocontrol action. These include the release of non‑ribosomal peptides (NRPs) and polyketides (PKs) with antimicrobial action, hormone production, colonization ability, biofilm formation and competition for space and nutrients, synthesis of acyl homoserine lactonases (AHSL), production of volatile organic compounds (VOCs), and induction of systemic resistance (ISR) (2525. Non‑ribosomal peptides produced by Bacillus species, such as iturins and phenazines among others, have shown antibacterial and antifungal activity, with effectiveness against Fusarium, Alternaria, Sclerotium, Cercospora, and Colletotrichum. Strains of B. siamensis showed activity against F. oxysporum forma specialis cubense (FOCTR4) (2626. Kulkova I, Dobrzyński J, Kowalczyk P, Bełźecki G, Kramkowski K. Available from: https://doi.org/10.3390/ijms24119759.), an effect associated with the production of a wide variety of siderophores by the bacterium (2727. Shen N, Li S, Li SY, Zhang H, Jiang M. The siderophore‑producing bacterium, Bacillus siamensis Gxun‑6, has an antifungal activity against Fusarium oxysporum and promotes the growth of banana. gypt J Biol Pest Control. 2022;32(1):34. On the other hand, the action of Bacillus strains against C. tuberculata was demonstrated, with mycelial inhibition percentage values close to 30 % (2828. Toledo-Hernández E, Peña-Chora G, Toribio-Jiménez J, Romero-Ramírez Y, Bolaños-Dircio A, Velázquez-del Valle MG, Hernández-Lauzardo AN, León-Rodríguez R, Silvana Vero S. These authors related the inhibitory effect to the production of volatile and diffusible metabolites by the bacteria.

Other studies have highlighted the potential of Bacillus for the control of Bipolaris sorokiniana, with a high in vitro inhibition percentage of the fungus, as well as the presence in this bacterium of genes associated with antagonistic activity related to the production of antibiotics, siderophores, and other substances (2424. The action of B. subtilis against Alternaria is also reported, with in vitro results superior to those achieved by commercial pesticides (2929. In addition to the fungi included in the study, genera such as Cladosporium and Corynespora have also been affected by strains belonging to the class Bacilli (1919. Rojas-Badía MM, Heydrich-Pérez M, Sánchez-Castro D, Tejera-Hernández B, Ríos-Rocafull Y, Lugo-Moya D. Bacterias del género Bacillus con potencialidades para la sostenibilidad agrícola en Cuba. Available from: http://www.revistaccuba.cu/index.php/revacc/article/view/1067. ), demonstrating their great potential for controlling fungal pathogens of high incidence in different plant species. Some of the substances mentioned above could have been released by the isolates selected in this research, which would explain their superior action compared to others that stood out in lytic enzyme production. Differences in the concentration and diversity of these metabolites can lead to the different results obtained for each of them.

Table 5 summarizes the main damage caused by the bacteria to the pathogens. Note that in some cases, the arrest of fungal growth was associated with elements such as mycelium coiling and abundant conidia production, which could constitute a selection criterion to enhance the biocontrol action of a product formulated from these bacteria.

Other research also reports the damage caused by Bacillus species on conidial germination, germ tube extension and morphology, and the plasma membrane of conidia and hyphae, leading to the death of phytopathogenic fungi such as Botrytis cinerea (3030. The role of compounds released by Bacillus species, particularly fengycins, in modifying the membrane permeability of fungal pathogens, including F. graminearum, is also highlighted (3131. In other studies, genera belonging to the class Bacilli have shown both indirect and direct mechanisms of plant growth promotion. Their potential to fix nitrogen and solubilize calcium, iron, and aluminum phosphate has been demonstrated, with solubilization indices between 1.30 and 2.59 (1919. Rojas-Badía MM, Heydrich-Pérez M, Sánchez-Castro D, Tejera-Hernández B, Ríos-Rocafull Y, Lugo-Moya D. Bacterias del género Bacillus con potencialidades para la sostenibilidad agrícola en Cuba. Added to this is the technological advantage of this group of bacteria due to their metabolic versatility, possessing structures that confer resistance under stress conditions, which would allow greater viability in the soil once applied (1414. Considering the aforementioned elements, it is a promising result to demonstrate that the microorganisms under study, associated with wheat cultivation, possess favorable characteristics for biological control, since these could counteract pathogen attack and stimulate plant growth through the application of bioproducts with an integral effect on the plant species (1919. Among the bacterial isolates, MTr2.1, MT3, and CTr2.1 stood out particularly for their significant action against the pathogens, as well as MH2, the only one that showed activity against four of the five fungal species. These bacteria are promising for future work assessing their potential as active ingredients in new biological controls for wheat cultivation in Cuba.

Conclusions

 

Bacteria associated with plants of the Cuban wheat cultivars 'Cuba C-204', 'I 399', and 'M04' can be found that have potential for the control of phytopathogenic fungi affecting this plant species. Particularly the isolates MH2, MTr2.1, MT3, and CTR2.1, selected for their antagonistic effect against Fusarium sp., Curvularia sp., Bipolaris sp., and Alternaria sp., are considered promising for the formulation of bioproducts aimed at the management of these pathogens in Cuba.
