Histology of adventitious shoot regeneration of leaves from in vitro cultured plantlets of guava (Psidium guajava L.), Cuban red dwarf cultivar

Abstract

A histological study was performed for
adventitious shoot regeneration of leaves from in vitro cultured
plantlets of guava (Psidium guajava L.), in order to determine
the morphogenetic mechanism that allows shoot regeneration
and to show the role of wound in this process. After an
adequate in vitro culture period, leaves were removed from
apical shoots and wounded at the main vein. Then, leaves
were inoculated in 25 mL of regeneration medium MS
supplemented with BAP 0.75 mg.L-1 and AIA 0.1 mg.L-1. The
explants were sampled with a frequency of 11 days during
44 days of culture. The samples were processed and stained
with Safranin-Fast Green technique. In different slides, it was
observed that the protuberances or �??little meristematic
nodules�?� formed have a pluri-tissular origin. In several cases,
meristematic cellular divisions began from epidermal and subepidermal
tissues, while in other cases, the vascular
parenchymatic tissues began to develop with continued
anticlinal and periclinal cell divisions until forming the nodular
structures. Also, it was demonstrated that the most important
morphogenic ability of guava leaves takes place at the main
vein and it is generally associated with the wounded region.
On the other hand, it was confirmed that protuberances are
nodular structures organized in meristematic centres which
achieve a high differentiation stage that allows independence
and the reorganization of a new caulinar meristem. With these
results, it was possible to verify that the adventitious
regenerated shoots from leaves of in vitro cultured plantlets
of guava have an indirect organogenic origin.
