A method for disinfecting and in vitro establishing mint (Mentha arvensis L.) explants

Abstract

The Mentha genus constitutes an important
source of raw material for several industries as perfumery,
cosmetics, distillery and pharmacy. In the state of Chihuahua,
Mexico, several species from this genus have been recently
introduced with commercial aims, and in vitro propagation
has been used with relative success in the massive
multiplication of one of them. The occurrence of endogenous
contaminations has been a problem for in vitro establishment
of these and some other commercial species. A method for
disinfection and in vitro establishment of explants from Mentha
arvensis L. is reported after a previous detection of two bacteria
genera (Streptobacillus and Enterobacter) abundantly
present in explants from this species. The method is based on
the combination of employing of chloramphenicol in a treatment
previous to implantation and as a constituent of the culture
medium,which allowed to obtain 65 % microorganism-free
explants. The causes for the presence of contamination in
explants from M. arvensis L. are discussed, and the method for
its application in other species of Mentha is recommended.
