Electrophoretic patterns of peroxidases, sterases, polyphenoloxidases and leaf damage in rice (Oryza sativa L.) seedlings under low temperature stress conditions

Abstract

This work was developed at Plant Genetics and
Breeding Laboratory from the National Institute of Agricultural
Sciences in 2002, with the objective to find electrophoretic
polymorphic bands that can be used as markers to select rice
genotypes with cold tolerance. The effect of different
temperature treatments was studied (0, 5, 10, 15 and 20oC) on
rice seedlings 25 days after germinating Dodo and IR-837,
resistant and susceptible varieties, respectively, which grew
under controlled conditions (25oC). The extraction was carried
out at three different moments and the isoenzimatic sterase,
peroxidase and polyphenoloxidase systems were studied, as
well as leaf damage measured by membrane permeability.
Results indicated that immediately after having applied
temperature stress, the most significant changes took place in
the electrophoretic patterns and it was detected that the most
sensitive system to temperature changes was
polyphenoloxidases. When reducing temperatures, leaf damage
percentage of resistant and susceptible varieties increased
gradually and abruptly, respectively.
