Effect of simulated drought with PEG-6000 on seedling germination and growth of two wheat varieties

Abstract
on wheat seedling germination and growth was evaluated after
selecting seeds from Cuba-C-204 and INIFAT RM-36 varieties.
The varieties were placed in petri dishes with three filter papers
and wet with 15 mL of a PEG-6000 solution at the following
osmotic pressures: 0, -0.3, -0.5, -0.75, -1.0 and -1.5 MPa. Petri
dishes were randomly distributed using 100 seeds per each
experimental treatment arranged in four repetitions. After seven
days, germination percentage (G) was determined, whereas
seedling height (AP), root length (LR) and the accumulation
of fresh (MF) and dry matters (MS) were evaluated at 15 days.
The linear regression equation was established for each variety
among the evaluated indicators, using the control percentage
and osmotic pressure levels as values that caused germination
and seedling growth reduction in 50 %. From the original
records, the inhibition was determined analyzing statistically
the results through the single variance analysis and Duncan�??s
test. Results showed that wheat germination and seedling
growth were affected by PEG simulated drought, it increasing
with the osmotic pressure of the solutions evaluated; therefore,
germination percentage and dry matter accumulation were the
most sensible parameters. The varieties presented a
differentiated response to the stress, INIFAT RM-36 being the
most tolerant variety considering germination and seedling
growth as an evaluation criterion, indicating the potential use
of these evaluations to select drought tolerant materials in the
early stages of a genetic program with this purpose.
