A new isolation and quantification approach of Collembola insects

Abstract

Recently, it has been reported that some members of the Collembola order feed from the external mycelia and propagules of arbuscular mycorrhizal fungi (AMF), they giving
rise mainly to negative effects on the development of symbiosis. Two experiments were designed with the objective of obtaining an optimum and efficient method enabling the isolation of Collembola in soils: the first one established a comparison between the commonly used methodology for animal isolation (Tullgren/Berlese funnels) and the traditional method of AMF spore count (wet sieving and decanting). The second experiment optimized the traditional approach achieving better results. A randomized complete design was used in both cases, the first one of simple classification and the second one
with bifactorial arrangement. Duncan´s test at 5 % was employed to compare treatment means. It was proved that
through wet sieving method, a larger number of individuals could be isolated, the best extractions being performed at a
centrifugation rate of 2 000 rpm and the longest time (five minutes). This method was more advantageous, not only for
significantly reducing the isolation time of individuals (five
minutes vs five days), but also because it allows to quantify
more alive individuals with an upper biological quality.
